Fibrinogen, prothrombin, thromboplastin, calcium, and antihemophilic factor are various types of blood clotting factors. Prothrombin is a plasma protein present in normal plasma. It is an unstable protein that can be split easily into thrombin. Prothrombin is continually formed by the liver.

Thrombin is a protein enzyme with weak proteolytic capabilities. It acts on fibrinogen to form one molecule of fibrin monomer. Fibrin monomers polymerize with one another to form fibrin fibers. Thrombin stimulates platelets to release adenosine diphosphate (ADP).

Coagulation testing is employed to determine unexplained bleeding. It is also used in pre- and perioperative testing, monitoring of anticoagulant treatment, and research. Prothrombin Time (PT), Activated Partial Thromboplastin Clotting Time (APTT), Thrombin Time (TT), and Activated Clotting Time (ACT) are various test types.

PT measures the time it takes plasma to clot when exposed to tissue factor and reflects the extrinsic and common pathways of coagulation.

APTT measures the time it takes plasma to clot when exposed to substances that activate the contact factors. ACT measures the time it takes whole blood (rather than plasma) to clot when exposed to an activator of the intrinsic pathway.
